目的:探讨肝肠-钙黏连蛋白(liver-intestine cadherin,CDH17)过表达对胃癌BGC-823细胞增殖、侵袭及迁移的影响。方法:采用脂质体转染法建立CDH17基因过表达的BGC-823细胞株;分别采用实时荧光定量-PCR(real-time fluorogenic quantitative-PCR,RFQ-PCR)和蛋白质印迹法检测转染重组表达载体pcDNA3.1(-)/CDH17后,BGC-823细胞中CDH17mRNA和蛋白的表达水平。MTT法检测CDH17基因过表达对BGC-823细胞增殖的影响;体外侵袭实验及划痕实验检测CDH17基因过表达对BGC-823细胞迁移及侵袭能力的改变。结果:RFQ-PCR及蛋白质印迹法检测结果显示,转染重组表达载体pcDNA3.1(-)/CDH17的BGC-823细胞中CDH17mRNA及蛋白均过表达;CDH17过表达能明显提高BGC-823细胞的增殖、侵袭及迁移能力(P<0.05)。结论:CDH17基因在BGC-823细胞中过表达能提高胃腺癌的增殖、侵袭及迁移能力,并促进胃腺癌的发生及发展。
Objective: To investigate the effect of liver-intestine cadherin (CDH17) overexpression on proliferation, invasion and migration of gastric cancer BGC-823 cells. METHODS: The BGC-823 cell line over-expressing CDH17 gene was established by liposome transfection, and the expression of the recombinant plasmid was detected by real-time fluorescence quantitative PCR (RFQ-PCR) and Western blotting respectively. Expression levels of CDH17 mRNA and protein in BGC-823 cells after vector pcDNA3.1(-)/CDH17. The effect of overexpression of CDH17 gene on the proliferation of BGC-823 cells was detected by MTT assay. The invasion and invasion of BGC-823 cells were detected by in vitro invasion assay and scratch assay. RESULTS: The results of RFQ-PCR and Western blotting showed that CDH17 mRNA and protein were over-expressed in BGC-823 cells transfected with the recombinant expression vector pcDNA3.1(-)/CDH17; overexpression of CDH17 could significantly increase the expression of BGC-823 cells. Proliferation, invasion and migration (P<0.05). Conclusion: Overexpression of CDH17 gene in BGC-823 cells can increase the proliferation, invasion and migration of gastric adenocarcinoma, and promote the occurrence and development of gastric adenocarcinoma.
