目的研究甘氨双唑钠(CMNa)在乏氧环境下对人胰腺癌Panc-1细胞的放射增敏效果。方法在乏氧和加不同浓度CMNa环境下对体外培养的人胰腺癌Panc-1细胞进行单次大剂量的γ射线照射,通过微孔板荧光酶标仪检测细胞死亡率来研究放射增敏作用。CMNa在乏氧环境下的选择性增敏效果利用流式细胞技术进行验证。结果胰腺癌Panc-1细胞30Gy照射后的细胞死亡率在乏氧环境下随CMNa浓度的增加显著增高;而在常规环境下CMNa却无明显的放射增敏作用。CMNa在乏氧环境下的选择性增敏作用具有时间相关性。结论CMNa在乏氧环境下对人胰腺癌细胞大剂量γ射线照射具有明显的增敏效果。
Objective To investigate the radiosensitization effect of glycidyridium sodium (CMNa) on human pancreatic cancer Panc-1 cells under hypoxic conditions. Methods Human pancreatic cancer Panc-1 cells cultured in vitro were exposed to γ-rays in a single dose under hypoxia and different concentrations of CMNa. The radiosensitization effect was studied by measuring the cell death rate by microplate fluorescence microplate reader . The selective sensitization of CMNa under hypoxic conditions was verified by flow cytometry. Results The cell death rate of Panc-1 cells after 30Gy irradiation was significantly increased with the increase of CMNa concentration in hypoxia environment. However, CMNa had no significant radiosensitization effect under normal conditions. The selective sensitization of CMNa under hypoxic conditions has a time-dependent effect. Conclusion CMNa can sensitize human pancreatic cancer cells to high dose γ-ray irradiation under hypoxia condition.
