目的:探讨表没食子儿茶素没食子酸酯(EGCG)对乳腺癌细胞MCF-7生长的影响及对乳腺癌细胞MDA-MB-231迁移的影响。方法:MCF-7细胞培养贴壁之后,加入EGCG处理,2d后收集蛋白,采用Western Blot检测磷酸化p38丝裂原活化蛋白激酶(phospho-p38MAPK)的表达;同样处理后收集活细胞,用细胞计数法检测细胞的存活;取对数生长期的MDA-MB-231细胞,分至6孔板培养,使用EGCG处理后,采用细胞划线法探测乳腺癌细胞的迁移。结果:使用EGCG处理乳腺癌细胞后,phospho-p38MAPK的表达降低,EGCG处理乳腺癌细胞4d后其增殖率降低50%,迁移活性降低。结论:EGCG处理乳腺癌细胞能抑制肿瘤细胞的生长以及迁移,这与p38 MAPK信号通路相关。
Objective: To investigate the effect of epigallocatechin-3-gallate (EGCG) on the growth of breast cancer cell line MCF-7 and its effect on the migration of breast cancer cell line MDA-MB-231. Methods: After adherent cells were treated with EGCG, the protein was collected 2 days later. The expression of phospho-p38MAPK protein was detected by Western Blot. After the same treatment, the cells were collected and treated with cells Counting method was used to test the survival of cells. MDA-MB-231 cells in logarithmic growth phase were cultured in 6-well plates and treated with EGCG to detect the migration of breast cancer cells. Results: After treated with EGCG, the expression of phospho-p38MAPK decreased. After 4 days, the proliferation rate of EGCG decreased by 50% and the migration activity decreased. Conclusion: EGCG treatment of breast cancer cells can inhibit tumor cell growth and migration, which is related to the p38 MAPK signaling pathway.
