目的 :分析当归多糖AP Ⅰ ,AP Ⅱ ,AP Ⅲ ,AP Ⅳ各组分的重均相对分子质量分布及其电荷特性 ;探讨多糖类生物大分子的组效关系研究方法。方法 :采用高效凝胶过滤色谱法、阴离子交换色谱法、二极管阵列检测器进行检测 ;改良苯酚 硫酸法测定糖分部 ;ServaBlue G染料结合法测定蛋白质分部。结果 :当归多糖AP Ⅰ ,AP Ⅱ ,AP Ⅲ ,AP Ⅳ分别由 4~ 5个具有不同重均相对分子质量分布及电荷特性的多糖组分组成 ;4种多糖中还分别含有一定量的游离或结合蛋白质。结论 :当归多糖AP Ⅰ ,AP Ⅱ ,AP Ⅲ ,AP Ⅳ为组成成分不同的多糖 ;该方法可为多糖组效关系研究提供简便、实用的分析方法。
Objective: To analyze the weight-average molecular weight distribution and charge characteristics of polysaccharides AP I, AP II, AP III and AP IV in Angelica sinensis. To explore the method for the study of the relationship between the polysaccharide biomacromolecules. Methods : High-performance gel filtration chromatography, anion exchange chromatography and diode array detector were used for detection. The improved phenol-sulfuric acid method was used to determine the sugar fraction. ServaBlue G dye binding method was used to determine the protein fraction. RESULTS: Angelica polysaccharides AP I, AP II, AP III and AP IV were composed of 4 to 5 polysaccharide components with different weight-average molecular weight distributions and charge characteristics. The four polysaccharides also contained a certain amount of free or Bind protein. Conclusion : Angelica polysaccharides AP I, AP II, AP III and AP IV are polysaccharides with different composition. This method can provide a simple and practical analysis method for the study of polysaccharide composition and efficacy.
