本试验利用二倍体栽培稻(Oryza sativa L.)品种8204,染色体组型为AA,2n=24作为母本,四倍体小粒野生稻(Oryza minuta)染色体组型为BBCC,2n=4x=48作父本杂交,获得异源三倍体杂交种,染色体组型为ABC.F_1根尖检查,染色体数目为2n=3X=36.用该异源三倍体杂交种的幼穗诱导愈伤组织,经长期继代培养进行原生质体游离,获得了再生植株白化苗.试验结果表明,用琼脂糖包埋法可促进原生质体再生愈伤组织.分化培养基用玉米素、激动素和6—卞基嘌呤三种细胞分裂素处理,6—卞基嘌呤表现出促进芽和根的分化的作用.基本培养基N_6比MS更为有效地促进植株再生而得到白化苗.本文旨在利用野生稻资源改良水稻栽培种的性状与培育新品种方法探索一条新的途径并为稻属种间细胞融合提供参考.
In this study, the diploid Oryza sativa cultivar (Oryza sativa L.) cultivar 8204 was used. The chromosome constitution was AA and 2n = 24 was used as the female parent. The chromosome constitution of Oryza minuta was BBCC, 2n = 4x = 48 crossed with the parents to obtain allotetraploid hybrids, chromosome type ABC.F_1 root tip examination, the number of chromosomes 2n = 3X = 36.
