用~(13)C NMR研究顺式二氯二氨合铂(简称顺铂)与鸟苷在中性介质中作用时,发现除生成N_7与铂配位的[Pt(NH_3)_2(N_7—Guo)_2]~(2+)外,尚生成鸟苷的N_7·N_1均与铂配位的[Pt(NH_3)_2(N_7·N_1—GuoH_(-1))]_n~(n+[1])。最近我们又成功地分离和析出了该化合物,经元素分析及~(13)C NMR谱表征为[{NH_3)_2Pt}_8(N_7·N_1—GuoH_(-1))_7(OH)_2](NO_3)_7~([2])。这些结果表明,顺铂不仅可能与癌细胞DNA形成N_7·N_7链内交联,亦可能形成N_7·N_1链内交联,从而抑制癌细胞中DNA的复制而具有抗癌活性。二种交联模型的CNDO/2研究又指出,N_7·N_7交联模型只能使G—C对(鸟嘌呤、胞嘧啶碱基对)间的氢键在一定程度上削弱,但不能使其全部
The results of ~ (13) C NMR studies showed that cisplatin (cisplatin) and guanosine (N_7 · N_1-GuoH_ (-1))] _n ~ (n + [1]) with the coordination of platinum with N_7 · N_1, ).
