目的:研究改构型酸性成纤维细胞生长因子(MaFGF)对顺铂(DDP)引起的体外培养的肾小管上皮细胞损害的保护作用。方法:将原代培养的肾小管上皮细胞接种于96孔培养板:(1)培养72h后加入一系列浓度的DDP,实验组在DDP作用12h后加入不同浓度MaFGF,再培养36h后用WST-8法检测细胞存活率。(2)以DDP建立损伤模型,并在加药后12h加入一定量的MaFGF,观察MaFGF对肾小管上皮细胞的保护作用。结果:(1)MaFGF能使DDP对肾小管上皮细胞的半数抑制浓度(IC50)升高。(2)DDP组与对照组比较,各生化和酶学指标差异均有统计学意义;而MaFGF+DDP组与对照组比较,SOD、GSH-Px酶活性差异无统计学意义,MDA、NO升高差异仍有统计学意义。结论:MaFGF对DDP损伤的肾小管上皮细胞有明显的保护作用。
AIM: To investigate the protective effect of modified acidic fibroblast growth factor (MaFGF) on renal tubular epithelial cells induced by cisplatin (DDP) in vitro. Methods: Primary cultured human renal tubular epithelial cells were inoculated into 96-well culture plates: (1) A series of concentrations of DDP were added after 72 hours of culture. The experimental groups were treated with different concentrations of MaFGF for 12 hours after DDP treatment. 8 method to detect cell survival rate. (2) The model of injury was established by DDP, and a certain amount of MaFGF was added 12 h after dosing to observe the protective effect of MaFGF on renal tubular epithelial cells. Results: (1) MaFGF increased the half maximal inhibitory concentration (IC50) of DDP on renal tubular epithelial cells. (2) Compared with the control group, the differences of biochemical and enzymological indexes in DDP group were statistically significant. However, the activity of SOD and GSH-Px in MaFGF + DDP group was not significantly different from that in control group High differences are still statistically significant. Conclusion: MaFGF has a significant protective effect on DDP-injured renal tubular epithelial cells.
