目的:探索简单高效的获取人脐静脉内皮细胞(HUVECs)的方法。方法:采用Ⅱ型胶原酶对健康产妇正常分娩的脐带进行消化,分离出HUVECs,予含10%胎牛血清、1%内皮细胞生长液的PRM 1640培养液,在37℃、5% CO_2条件下培养,分别进行细胞鉴定及活力检测。结果:分离获取的HUVECs呈典型的“鹅卵石、铺路石”样,有特异性vWF抗原表达,细胞活力好。结论:Ⅱ型胶原酶对健康产妇正常分娩的脐带进行消化分离,可获取大量纯度高、细胞活力好的HUVECs,简单高效、可行性强。
Objective: To explore a simple and efficient method for obtaining human umbilical vein endothelial cells (HUVECs). Methods: Human umbilical cord was digested with type Ⅱ collagenase to produce normal pregnant women. HUVECs were isolated and cultured in PRM 1640 medium containing 10% fetal bovine serum and 1% endothelial cell growth medium. Under the conditions of 37 ℃ and 5% CO 2 Culture, respectively, cell identification and vitality testing. Results: The isolated HUVECs were typical “cobblestone, paving stone” samples, with specific vWF antigen expression, good cell viability. CONCLUSION: Type Ⅱ collagenase digests the umbilical cord of normal delivery of healthy maternal, and can obtain a large number of HUVECs with high purity and good cell viability. It is simple, efficient and feasible.
