目的 比较骨髓与脐带血细胞体外培养基质细胞的基本特性,为基质细胞的选择应用提供依据。方法用Dexter长期培养体系培养骨髓和脐带血基质细胞,以细胞增殖、细胞形态、细胞化学染色、细胞表面抗原及基质细胞支持另一骨髓细胞形成的鹅卵石造血区、长期培养起始细胞为指标,比较两者的生长特性、成分及功能。结果生长特性:出现贴壁细胞时间,骨髓细胞为培养3 d,脐带血细胞为培养5~6 d;细胞融合成片时间,骨髓细胞为培养10~14 d,脐带血细胞为培养12~18 d;第21天细胞增殖数,骨髓比脐带血细胞增殖少。细胞成分:21d培养后细胞成分,骨髓来源者以成纤维细胞为主,其次是巨噬细胞与内皮细胞,脂肪细胞最少;而脐带血细胞来源者,以巨噬细胞为主,其次是内皮细胞、成纤维细胞,偶见脂肪细胞;细胞化学与上述结果基本一致;细胞表面抗原检测,CD14、CD45的表达骨髓细胞明显低于脐带血细胞。细胞功能:骨髓来源的基质细胞较脐带血细胞的基质细胞支持另一骨髓细胞形成的鹅卵石造血区和长期培养起始细胞明显多。结论①生长特性:形成贴壁细胞时间骨髓较脐带血短,骨髓细胞比脐带血细胞有核细胞数增殖快、持续时间相对短。②细胞成分特性:骨髓来源形成的基质细胞以成纤维细胞为主,脐带血来源者以巨噬细胞为主。③细胞功能特性:骨髓细 Objective To compare the basic characteristics of stromal cells cultured in vitro with bone marrow and umbilical cord blood cells, and to provide basis for the selection and application of stromal cells. Methods Bone marrow and umbilical cord blood stromal cells were cultured with Dexter long-term culture system. The long-term culture of the starting cells was used as an indicator of cell proliferation, cell morphology, cytochemical staining, cell surface antigens and stromal cells supporting pebble hematopoietic cells formed by another bone marrow cells. Compare the growth characteristics, composition and function of the two. Results The growth characteristics were as follows: the adherent cells appeared for 3 days, the cord blood cells were cultured for 5 to 6 days, the time for cell fusion into tablets was 10-14 days for bone marrow cells and 12-18 days for cord blood cells. The number of cell proliferation on day 21, bone marrow less than cord blood cell proliferation. Cell components: After 21 days of culture, the cell components were mainly fibroblasts, followed by macrophages and endothelial cells, with the smallest number of adipocytes; while the umbilical cord blood cells were mainly macrophages, followed by endothelial cells, Fibroblasts, occasionally adipocytes; cytochemistry and the above results are basically the same; cell surface antigen detection, CD14, CD45 expression in bone marrow cells was significantly lower than umbilical cord blood cells. Cell function: Bone marrow-derived stromal cells have significantly more pebble hematopoietic cells and long-term culture initiating cells than stromal cells of cord blood cells that support another bone marrow cell. Conclusion ① growth characteristics: the formation of adherent cells in bone marrow than cord blood short, bone marrow cells than cord blood cells nucleated cells proliferate faster, the duration is relatively short. ② cell component characteristics: the formation of bone marrow stromal cells to fibroblasts, umbilical cord blood-derived macrophages. ③ cell function features: fine bone marrow
