背景:研究发现脑缺血后多形核白细胞(polymorphonuclearleukocyte,PMNL)与微血管内皮细胞(capillaryendothelialcell,CEC)间的黏附增多,应用抗细胞间黏附分子-1(intercellularadhesionmolecule-1,I-CAM-1)抗体可减轻神经元的缺血性损伤。但是目前还没有直接的证据表明抗-ICAM-1抗体可以抑制PMNL与CEC间的黏附过程。目的:观察脑缺血再灌注后PMNL与CEC间的黏附性变化,探讨应用抗-ICAM-1抗体对脑缺血再灌注后PMNL与CEC间黏附性的影响。设计:完全随机设计。地点和对象:实验地点设在第三军医大学大坪医院神经内科实验室。以成年雄性Wistar大鼠作为实验动物,分为对照组(n=4)、假手术组(n=6)、脑缺血再灌注组(n=10)和治疗组(n=10)。干预:假手术组、脑缺血再灌注组和治疗组大鼠分别于假手术和再灌注4,12,24h后抽取静脉血2~4mL。治疗组大鼠于缺血再灌注后1h静脉注射抗ICAM-1抗体(1mg/kg)。用右旋糖酐沉淀法和Percoll密度梯度离心法分离出PMNL,加入培养的CEC,1h后进行微管吸吮检测。主要观察指标:PMNL与CEC间的黏附力和黏附应力。结果:脑缺血再灌注4h后PMNL和CEC间的黏附力和黏附应力明显升高,并于12h达到高峰。治疗组大鼠在各时间点的黏附力犤4h:(14.3±0.6)N,12h:(16.2±0.8)N,24h:(13.7±0.5)N犦和黏附应力犤4
BACKGROUND: The study found that the adhesion between polymorphonuclear leukocyte (PMNL) and capillary endothelial cell (CEC) increased after focal cerebral ischemia. Intercellular adhesion molecule-1 (I-CAM- Antibodies reduce ischemic damage in neurons. However, there is no direct evidence that anti-ICAM-1 antibody can inhibit the adhesion between PMNL and CEC. Objective: To observe the changes of adhesion between PMNL and CEC after cerebral ischemia-reperfusion and to investigate the effect of anti-ICAM-1 antibody on the adhesion between PMNL and CEC after cerebral ischemia-reperfusion. Design: completely random design. Location and object: The experimental site is located in Daping Hospital Neurology Laboratory, Third Military Medical University. The adult male Wistar rats were divided into control group (n = 4), sham operation group (n = 6), cerebral ischemia reperfusion group (n = 10) and treatment group (n = 10). Intervention: sham operation group, cerebral ischemia reperfusion group and treatment group rats were sham operated and reperfusion 4,12,24h after venous blood drawn 2 ~ 4mL. The rats in the treatment group were injected with anti-ICAM-1 antibody (1mg / kg) intravenously 1h after ischemia-reperfusion. PMNL was isolated by dextran precipitation and Percoll density gradient centrifugation, and cultured in CEC. Microtubule sucking was performed after 1 h. MAIN OUTCOME MEASURES: Adhesion and adhesion stress between PMNL and CEC. Results: The adhesion and adhesive stress between PMNL and CEC increased significantly after cerebral ischemic reperfusion 4h, reaching the peak at 12h.
