目的:探讨心脑通络液对动脉粥样硬化兔平滑肌细胞的干预作用。方法:选取健康雄性新西兰大耳兔40只,随机分成假手术组、模型组、血脂康组、心脑通络液高、低剂量组,对除假手术组外的其余组动物建立腹主动脉粥样硬化模型。造模成功后,各组分别灌胃给药,连续治疗8周,采用SP法测定各组家兔Bcl-2、Caspase-3的阳性表达率。结果:与假手术组比较,模型组Bcl-2含量明显降低,Casapase-3含量明显升高,差异有统计学意义(P<0.05);与模型组比较,血脂康组和高低剂量组Bcl-2含量均增高、Caspase-3含量均降低(P<0.05),以高剂量改善最显著(P<0.05)。结论:心脑通络液可能通过对bcl-2及Caspase-3蛋白表达的调控而降低动脉斑块内过度凋亡的平滑肌细胞,从而抑制粥样斑块的形成。
Objective: To explore the intervention of Xinnaotongluo liquid on the smooth muscle cells of atherosclerosis rabbits. Methods: Forty male New Zealand white rabbits were randomly divided into sham operation group, model group, Xuezhikang group and Xinnaotongluo liquid high and low dose groups. The animals in the other groups except the sham operation group were established abdominal aorta Atherosclerosis model. After successful modeling, each group was administered by gavage for 8 weeks continuously, and the positive rate of Bcl-2 and Caspase-3 in rabbits in each group was determined by SP method. Results: Compared with the sham operation group, Bcl-2 content in model group decreased significantly and Caspase-3 content increased significantly (P <0.05). Compared with model group, Bcl- 2 content increased, Caspase-3 content decreased (P <0.05), to improve the most significant dose (P <0.05). Conclusion: Xinnaotongluo Decoction can inhibit the formation of atherosclerotic plaque by reducing the excessive apoptosis of smooth muscle cells in arterial plaque through the regulation of bcl-2 and Caspase-3 protein expression.
