在水稻雄性不育系花粉细胞败育程度鉴定方法上,传统的单纯显微图像分析方法或纯光谱分析方法存在定性、定位、定量和定时四位一体分析能力不足的问题。本文在普通荧光显微镜的基础上,研制出一种基于LCTF(Liquid Crystal Tunable Filter,液晶可调谐滤光器)的显微光谱成像分析系统,并将其引入到水稻花粉发育过程中的细胞学定量研究,初步建立起一套针对水稻花粉透射光光谱图像分析和荧光光谱图像分析新方法。与传统的纯光谱或纯图像分析方法相比,本分析方法具有独特的定性、定量、定位和定时四位一体的分析能力,其对水稻雄性不育系花粉败育程度的鉴定将更为准确。本方法对于其它植物和动物细胞的定量分析同样具有借鉴和推广意义。
In the method of identification of pollen cell abortion in male sterile lines of rice, the traditional methods of pure microscopic image analysis or pure spectrum analysis have the problems of lack of capacity of qualitative, localization, quantitative and time-keeping analysis. In this paper, based on the ordinary fluorescence microscope, a LCTF (Liquid Crystal Tunable Filter) based microspectroscopic imaging analysis system was developed and introduced into the cytology of pollen development in rice Research, initially set up a set of new methods for rice pollen transmission light spectrum image analysis and fluorescence spectroscopy image analysis. Compared with the traditional methods of pure spectrum or pure image analysis, this method has a unique ability to analyze, quantify, locate and quantify tetracyclic genes, and it will be more accurate to identify the degree of pollen abortion in male sterile lines of rice . The method for quantitative analysis of other plant and animal cells also has reference and promotion of significance.
