目的:了解豚鼠变应性鼻炎动物模型的鼻黏膜中诱导型NOS(iNOS)和内皮型NOS(eNOS)的表达、分布及地氯雷他定对其的影响。方法:实验豚鼠36只,随机分为模型组、治疗组和阴性对照组,经抗原致敏激发后,获取鼻黏膜组织,进行免疫组织化学染色,片将切片进行半定量图像分析。结果:eNOS的表达在治疗组和模型组均高于阴性对照组(分别P<0.05和P<0.01);治疗组和模型组比较无统计学意义。iNOS的表达在各组间的比较无统计学意义。eNOS除了iNOS的分布部位外,在杯状细胞中也有表达。结论:在豚鼠的变应性鼻炎模型中.eNOS可能比iNOS具有更重要的调节NO水平的功能;地氯雷他定对iNOS和eNOS的表达无影响。
Objective: To investigate the expression and distribution of inducible NOS (iNOS) and endothelial NOS (eNOS) in nasal mucosa of guinea pig allergic rhinitis animal model and the effect of desloratadine on it. Methods: Thirty-six guinea pigs were randomly divided into model group, treatment group and negative control group. After sensitized by antigen, nasal mucosa tissues were harvested for immunohistochemical staining. Semi-quantitative image analysis was performed on the slices. Results: The expression of eNOS in the treatment group and the model group was higher than that in the negative control group (P <0.05 and P <0.01, respectively). There was no significant difference between the treatment group and the model group. The expression of iNOS was not statistically significant among the groups. In addition to the distribution of eNOS iNOS, also expressed in goblet cells. CONCLUSIONS: In NSC models of guinea pigs, eNOS may have a more important role in regulating NO than iNOS; desloratadine has no effect on the expression of iNOS and eNOS.
