目的 :探讨流式细胞仪 (FCM)在膀胱癌多药耐受研究中的应用。方法 :利用蛋白质印迹、免疫细胞化学和 FCM检测并比较膀胱癌多药耐受细胞 EJ/MRP的多药耐受糖蛋白 (P- gp)和多药耐受相关蛋白 (MRP)的表达 ;利用 FCM检测了 EJ/MRP药物耐受的功能水平。结果 :EJ/MRP高表达 MRP,无明显 P- gp表达 ,FCM的检测结果与蛋白质印迹、免疫细胞化学的结果一致 ,但检测更简便 ;功能检测显示柔红霉素 (DNR)可以作为 FCM检测 MRP功能的发光探针 ,细胞外排药物的功能增强至少是 MRP高表达的膀胱癌细胞内药物聚集减少的一个重要原因。结论 :FCM是检测膀胱癌多药耐受蛋白质表达水平和 MRP功能水平的好方法 ,DNR是 FCM检测MRP功能的良好探针。
Objective: To investigate the application of flow cytometry (FCM) in multidrug resistance of bladder cancer. Methods: Western blotting, immunocytochemistry and FCM were used to detect and compare the expression of multi-drug resistant protein (Pgp) and multidrug resistance associated protein (MRP) in bladder cancer multidrug resistant EJ / MRP cells. FCM tested the functional level of EJ / MRP drug resistance. Results: MR / MRP expression of MRP, no significant expression of p-gp, FCM results and western blot, immunocytochemistry results, but the test more simple; functional test showed that daunorubicin (DNR) can be used as FCM detection MRP function of the luminescent probe, cell extracellular drug enhancements is at least MRP high expression of bladder cancer cells in a reduction in drug aggregation is an important reason. Conclusion: FCM is a good method to detect multidrug resistance protein expression and MRP function in bladder cancer. DNR is a good probe for detecting MRP function by FCM.
