目的:对板栗花粉中的甾醇类物质进行提取、分离与成分鉴定,为花粉的开发奠定理论基础。方法:以乙醇为提取剂,提取温度70℃,提取时间为1.5h,固液比1:10对板栗花粉中的植物甾醇进行了提取,并用AB-8大孔树脂对其进行纯化,洗脱体系为80mL60%乙醇、60mL丙酮。使用N-甲基-N-(三甲基硅烷基)三氟乙酰胺活化物Ⅱ(MSTFAactivatedⅡ)为衍生化试剂,衍生后注入气相色谱-质谱联用仪器进行分析。研究结果显示得到了5种匹配度较高的甾醇,分别是:胆固醇、(3β,24R)-麦角甾-5-烯-3-醇、豆甾醇、β-谷甾醇和(3β)-豆甾-5,24(28)-双烯-3-醇。另使用溶剂提取法提取雌二醇,并用酶联免疫法(ELISA)对板栗花粉中雌二醇的含量进行了研究。结果显示雌二醇最大提取量为261.08pg/g。
OBJECTIVE: To extract, isolate and identify sterols in chestnut pollen and lay a theoretical foundation for the development of pollen. Methods: The phytosterols in the chestnut pollen were extracted with ethanol as extraction solvent, extraction temperature was 70 ℃, extraction time was 1.5h and solid-liquid ratio was 1:10. The phytosterols were extracted from the chestnut pollen by using AB-8 macroporous resin System 80mL60% ethanol, 60mL acetone. Using N-methyl-N- (trimethylsilyl) trifluoroacetamide activator II (MSTFAactivatedⅡ) as a derivatization reagent, derivatization was performed and analyzed by gas chromatography-mass spectrometry. The results showed that five kinds of sterols with high matching degree were obtained: cholesterol, (3β, 24R) -eritost-5-en-3-ol, stigmasterol, β-sitosterol and -5,24 (28) -dien-3-ol. In addition, the solvent extraction method was used to extract estradiol, and the content of estradiol in chestnut pollen was studied by enzyme-linked immunosorbent assay (ELISA). The results showed that the maximum amount of estradiol extraction was 261.08 pg / g.
