目的:观察不同引经药配伍大黄-丹参药对抗大鼠肝纤维化的效应差异,探讨引经药的增效作用。方法:健康雄性SD大鼠60只,随机分为正常组,模型组,丹参-大黄药对组(5.0 g·kg~(-1)),药对加桔梗组(5.0 g·kg~(-1)+1.5 g·kg~(-1)),药对加柴胡组(5.0 g·kg~(-1)+1.5 g·kg~(-1))和药对加细辛组(5.0 g·kg~(-1)+0.3 g·kg~(-1)),除正常组外,其余各组均腹腔注射四氯化碳(CCl4)复制肝纤维化模型。从第3周开始,除注射造模外,各实验组分别给予相应药物灌胃,正常组、模型组灌胃双蒸水,连续6周。末次给药后,取血和肝脏,检测血清中丙氨酸氨基转移酶(ALT),天门冬氨酸氨基转移酶(AST),碱性磷酸酶(ALP)的活性;酶联免疫吸附法(ELISA)测定血清中层黏连蛋白(LN),透明质酸(HA)和Ⅲ型前胶原(PCⅢ)的含量;酸水解法检测肝匀浆中羟脯胺酸(HYP)的含量;观察肝脏的组织形态改变;免疫组化法检测肝组织中转化生长因子-β1(TGF-β1)的表达。结果:与模型组比较,各给药组大鼠血清中ALT,AST,ALP活性和LN,HA,PCⅢ的含量显著降低,肝匀浆中HYP含量降低,肝组织TGF-β1表达减少(P<0.05,P<0.01),肝组织结构明显改善;与药对组相比,药对加柴胡组大鼠ALT,AST,ALP活性和LN,HA,PCⅢ的含量降低,肝组织TGF-β1表达减少(P<0.05,P<0.01)。结论:大黄-丹参药对能有效减轻肝纤维化的程度,引经药柴胡可增强大黄-丹参药对的抗肝纤维化作用。
OBJECTIVE: To observe the difference of effect of Rhubarb - Salvia Miltiorrhiza on compatibility of rat liver fibrosis with different traditional Chinese medicines and to explore the synergistic effect of Danshen Medicine. Methods: Sixty healthy male Sprague Dawley rats were randomly divided into normal group, model group, Salviae Miltiorrhizae rhubarb group (5.0 g · kg -1) 1) and 1.5 g · kg ~ (-1)), respectively. The effects of the two drugs on gavage (5.0 g · kg -1 ± 1.5 g · kg -1) g · kg -1 and0.3 g · kg -1, respectively. Except the normal group, the other groups were intraperitoneally injected CCl 4 to replicate the hepatic fibrosis model. From the third week onwards, in addition to the injection molding, the experimental groups were given the corresponding drug gavage, normal group, model group gavage double distilled water for 6 weeks. After the last administration, blood and liver were taken for detection of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP) activity; enzyme-linked immunosorbent assay ELISA was used to detect the levels of serum laminin (LN), hyaluronic acid (HA) and procollagen III (Ⅲ) in the liver homogenates. The contents of hydroxyproline (HYP) in liver homogenates were detected by acid hydrolysis method. The morphological changes were observed. The expression of transforming growth factor-β1 (TGF-β1) in liver tissues was detected by immunohistochemistry. Results: Compared with the model group, the contents of ALT, AST, ALP, LN, HA and PCⅢ in the serum of rats in each group were significantly decreased, the content of HYP in liver homogenate decreased and the expression of TGF-β1 in liver decreased (P < 0.05, P <0.01). The structure of hepatic tissue was significantly improved. Compared with the drug group, the activity of ALT, AST, ALP and the content of LN, HA and PCⅢ were decreased in the Jia Chaochu group and the expression of TGF- Decrease (P <0.05, P <0.01). Conclusion: Rhubarb - Salvia Miltiorrhiza can effectively reduce the degree of liver fibrosis, the introduction of medicine Bupleurum can enhance rhubarb - Danshen on anti-liver fibrosis.
