目的:建立测定人血浆中阿奇霉素浓度的方法。方法:采用直接沉淀法处理血浆后以超高效液相色谱-串联质谱(UPLC-MS)法进样测定。色谱柱为Waters Acquity UPLC BEH C18流动相为乙腈-10 mmo·lmL-1甲酸铵水溶液(66∶34),流速为0.2 mL·min-1;正离子多离子反应监测(MRM)扫描分析,离子通道分别为m/z 748.36→591.10(阿奇霉素)、m/z 837.34→158.06(罗红霉素)。结果:阿奇霉素血药浓度在2~1 000 ng·mL-1范围内线性关系良好(r=0.999 5),定量下限为2 ng·mL-1平均方法回收率为102.6%~108.0%,平均提取回收率为93.2%~102.2%,日内、日间RSD均<8%。结论:本方法简便、快速、可靠、特异性强、灵敏度高,可用于阿奇霉素的药动学研究。
Objective: To establish a method for the determination of azithromycin in human plasma. Methods: The plasma was precipitated by direct precipitation method and then determined by ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS). The column was Waters Acquity UPLC BEH C18 with a mobile phase of acetonitrile-10 mmo · mL-1 ammonium formate (66:34) and a flow rate of 0.2 mL · min -1. The positive ion MRM scan analysis, Channels were m / z 748.36 → 591.10 (azithromycin), m / z 837.34 → 158.06 (roxithromycin). Results: The linearity of azithromycin in the range of 2 ~ 1 000 ng · mL-1 was good (r = 0.999 5), and the lower limit of quantification was 2 ng · mL-1. The average recoveries were 102.6% -108.0% The recoveries ranged from 93.2% to 102.2%, with daily and intraday RSDs <8%. Conclusion: The method is simple, rapid, reliable, specific, sensitive and can be used for the pharmacokinetics of azithromycin.
