目的分析乳腺癌细胞雌激素依赖性与畸胎瘤细胞源性生长因子(PC-cell derivedgrowth factor,PCDGF)表达之间的关系,探讨雌激素受体(ER)阴性乳腺癌患者接受内分泌治疗的可能性。方法荧光定量聚合酶链反应检测乳腺癌细胞系 MCF-7、MDA-MB-231、T47D、MDA-MB-435s中 PCDGF 的表达,CCK-8法检测不同浓度雌激素培养条件下细胞的存活情况。以 ER 阳性乳腺癌细胞系 MCF-7为对照,用 RNA 干扰技术抑制 ER 阴性乳腺癌细胞系 MDA-MB-231中 PCDGF 基因的表达,观察抑制前后雌激素依赖性的变化。结果 PCDGF 在4株乳腺癌细胞系中均有不同程度的表达,其中在 MDA-MB-435s 中表达量最高,PCDGF 高表达的乳腺癌细胞系的雌激素依赖性较高,PCDGF shRNA 可以有效抑制乳腺癌细胞系 MCF-7和 MDA-MB-213中 PCDGF 的表达(抑制率分别为81.1%和86.7%)。MDA-MB-231细胞系雌激素依赖性增加的程度高于 MCF-7细胞系(P<0.05)。结论 PCDGF 高表达于 ER 阴性乳腺癌细胞,抑制其表达可能会更好的逆转内分泌治疗的耐药性。
Objective To analyze the relationship between estrogen-dependent breast cancer cells and the expression of PC-derived derived growth factor (PCDGF), and to explore the possibility of endocrine therapy in estrogen receptor-negative breast cancer patients Sex. Methods The expression of PCDGF in breast cancer cell lines MCF-7, MDA-MB-231, T47D and MDA-MB-435s was detected by fluorescence quantitative polymerase chain reaction. The survival rate of cells under different concentrations of estrogen was detected by CCK- . Using ER-positive breast cancer cell line MCF-7 as control, the expression of PCDGF gene in ER-negative breast cancer cell line MDA-MB-231 was inhibited by RNA interference and the estrogen-dependent changes before and after inhibition were observed. Results PCDGF was expressed in all four breast cancer cell lines to varying degrees. Among them, the expression level of PCDGF was the highest in MDA-MB-435s and estrogen-dependent in PCDGF-overexpressing breast cancer cell lines. PCDGF shRNA could effectively inhibit PCDGF expression in breast cancer cell lines MCF-7 and MDA-MB-213 (81.1% and 86.7% inhibition, respectively). The estrogen-dependent increase of MDA-MB-231 cell line was higher than that of MCF-7 cell line (P <0.05). Conclusions PCDGF is overexpressed in ER-negative breast cancer cells. Suppressing the expression of PCDGF may reverse the resistance of endocrine therapy better.
