目的 探讨氧化砷(As2O3)对结肠癌细胞SW480的增殖抑制作用。方法 应用台盼蓝拒染法、MTT比色法及细胞克隆形成试验,研究As2O3对结肠癌细胞的增殖抑制和细胞毒作用。结果 As2O3能显著抑制结肠癌细胞的生长,5μmol/LAs2O3抑制强度明显强于1.25mol/L、1.25μmol/L和5μmol/L As2O3作用后克隆形成率分别为(9.6±2.1)%和(1.7±0.9)%,明显低于对照组的(48.2±6.7)%(P<0.01)。As2O3对结肠癌细胞有较强的细胞毒作用,且呈剂量和时间依赖性,其对结肠癌细胞的杀伤率显著高于5-氟尿嘧啶(5-FU)。As2O3与5-FU存在协同效应,联合应用后,对结肠癌细胞的杀伤率明显升高。结论 As2O3具有明显抑制结肠癌细胞增殖的作用,存在治疗结肠癌的潜在价值。
Objective To investigate the inhibitory effect of arsenic trioxide (As2O3) on the proliferation of colon cancer cell line SW480. Methods Trypan blue exclusion, MTT assay and cell clone formation assay were used to study the inhibitory effects of As2O3 on colon cancer cell proliferation and cytotoxicity. Results As2O3 could significantly inhibit the growth of colon cancer cells. The inhibitory concentration of 5μmol / L As2O3 was significantly stronger than that of 1.25mol / L, 1.25μmol / L and 5μmol / L As2O3 (9.6 ± 2.1)% and 0.9%), which was significantly lower than that of the control group (48.2 ± 6.7%) (P <0.01). As2O3 has a strong cytotoxic effect on colon cancer cells in a dose- and time-dependent manner. The cytotoxicity of As2O3 on colon cancer cells was significantly higher than that of 5-fluorouracil (5-FU). As2O3 and 5-FU synergistic effect exists, combined application, the killing rate of colon cancer cells was significantly increased. Conclusion As2O3 has a significant inhibitory effect on the proliferation of colon cancer cells, and has the potential value of treating colon cancer.
