目的 探讨输血后不同时间、不同输血量供者STRs基因型对受者STRs基因型的影响。方法 采集3例患者输血前以及输血后4 h、8h、12 h的血液和供者血液,EDTA抗凝,用Chelex- 10 0法提取DNA,选D1S5 4 9、D18S86 5、D3S175 4、D12 S391、D12 S375、D6 S4 77等6个STRs基因座进行PCR扩增,扩增产物用聚丙烯酰胺凝胶垂直板电泳,银染显色分型。结果 6个STRs基因座的检测结果显示受者输血前及输血后12 h内STRs基因型一致。结论 输血量不大于12 0 0 ml的受者输血后12 h内STRs基因型未受供者血液STRs基因型的影响。
Objective To investigate the effect of different STRs genotypes on the STRs genotypes of recipients at different times after transfusion. Methods Blood was collected from 3 patients before transfusion and after 4 h, 8 h, 12 h after blood transfusion. Blood and donor blood were collected and EDTA was anticoagulated. DNA was extracted by Chelex-10 method and selected for D1S5 4 9, D18S86 5, D3S175 4, D12 S391 , D12 S375, D6 S4 77 and other 6 STRs loci were amplified by PCR, and the amplified products were electrophoresed on polyacrylamide gel by vertical plate and silver staining. Results The results of six STRs loci showed that the genotypes of STRs were consistent before and 12 h after transfusion. Conclusion The genotypes of STRs were not affected by the STRs genotype of donor blood within 12 hours after blood transfusion.
