用全基因组微阵列比较了根瘤中苜蓿根瘤菌1021的nifA突变菌和野生菌的基因表达谱.分析结果表明,nifA的缺失引起601个基因的表达发生变化.这些基因分别属于生物大分子的合成代谢、三羧酸循环及呼吸代谢以及结瘤固氮过程等多个功能组,预示着根瘤中细菌的生长状态发生了显著的变化.在根瘤中,fixK以及受其激活的基因在nifA突变菌中的表达量显著高于野生菌.定量实时PCR分析表明,根瘤中fixLJ的转录水平在nifA突变菌中显著高于野生菌.通过统计学方法从13个表达发生显著变化的基因的上游调控序列中找到推测的NifA结合位点.
Genomic microarrays were used to compare the gene expression profiles of nifA and wild strains of nodulated alfalfa nodules 1021.
