目的探讨EB病毒(EBV)感染的系统性红斑狼疮(SLE)患者EBV基因BYRF1和BcLF1表达与SLE的关系。方法应用聚合酶链反应(PCR)-Southern杂交技术检测44例SLE患者和43名正常对照外周血单个核细胞中EB病毒(EBV)特异性DNA片段,反向(RT)-PCR检测EBV特异性DNA片段阳性患者病毒潜伏期基因BYRF1和增殖期基因BcLF1的表达。结果SLE患者组EBV特异性DNA片段BamH-ΙW阳性率为72.73%(32/44),明显高于正常对照组6.98%(3/43)(2χ=39.177 9,P=0)。32例EBV BamH-ΙW片段阳性标本中有20例BYRF1mRNA阳性,其中活动期患者9例,稳定期患者11例,活动期和稳定期患者阳性率的差异无显著性(P=0.290 7)。EBV增殖期基因BcLF1 mRNA 16例阳性,其中活动期患者14例,稳定期患者2例,活动期和稳定期患者之间的差异有显著性(P=0.000 2)。结论BYRF1 mRNA和BcLF1 mRNA的表达可能在一定程度上参与SLE的发病和病程进展。
Objective To investigate the relationship between EBV gene expression of BYRF1 and BcLF1 and SLE in patients with Epstein-Barr virus (EBV) infected systemic lupus erythematosus (SLE). Methods Epstein-Barr virus (EBV) specific DNA fragments were detected in 44 SLE patients and 43 normal control PBMCs by polymerase chain reaction-Southern blotting. The specificity of EBV was detected by RT-PCR Expression of BYRF1 and BcLF1 genes in DNA fragment positive patients. Results The positive rate of EBV-specific DNA fragment BamH-IW in SLE patients was 72.73% (32/44), which was significantly higher than that in normal controls (3/8) (2χ = 39.177 9, P = 0). There were 20 cases of positive BYRF1 mRNA in 32 cases of EBV BamH-1W fragment, including 9 cases of active stage and 11 cases of stable stage. There was no significant difference in the positive rate between active stage and stable stage (P = 0.2907). 16 cases of EBV proliferative phase gene BcLF1 mRNA were positive, including 14 active patients and 2 stable patients. The difference between active and stable patients was significant (P = 0.000 2). Conclusion The expression of BYRF1 mRNA and BcLF1 mRNA may be involved in the pathogenesis and progression of SLE to a certain extent.
