目的对河北省2007~2008年急性弛缓性麻痹(Acute Flaccid Paralysis,AFP)病例和密切接触者的粪便标本中,分离的脊髓灰质炎(脊灰)病毒进行VP1编码区基因特征分析,为维持无脊灰提供依据。方法按世界卫生组织《脊灰实验室手册》进行病毒分离与血清定型,用逆转录-聚合酶链反应扩增VP1编码区基因片段,并进行核苷酸序列测定和分析,构建基因亲缘关系树。结果河北省2007~2008年,从AFP病例和密切接触者粪便标本中共分离到脊灰病毒42株,以Ⅱ、Ⅲ型血清型为主,均为疫苗类似株,未发现野病毒和疫苗衍生脊灰病毒。结论脊灰病毒VP1编码区基因特征分析可用于识别脊灰病毒传播途径,并对免疫规划工作具有指导作用。
Objective To analyze the genetic characteristics of the VP1 coding region of poliomyelitis (poliovirus) isolated from acute flaccid paralysis (AFP) cases and close contact stool samples from 2007 to 2008 in Hebei Province. Polio provides the basis. Methods Virus isolation and serum typing were performed according to the “Polio Laboratory Manual” of the World Health Organization. The VP1 coding region gene fragment was amplified by reverse transcription - polymerase chain reaction (PCR) and the nucleotide sequence was determined and analyzed. Gene phylogenetic tree . Results From 2007 to 2008, 42 strains of poliovirus were isolated from the AFP cases and the close contact stool samples of Hebei Province. The serotypes Ⅱ and Ⅲ were all vaccine-like strains. No wild-virus and vaccine-derived ridges were found Gray virus. Conclusion The gene characteristic analysis of VP1 coding region of poliovirus can be used to identify the route of transmission of poliovirus and to guide the immunization planning.
