目的:研究定心胶囊对ACE2-/y小鼠血压升高和心功能异常的作用及其调控机制。方法:应用血管紧张素Ⅱ(Ang Ⅱ)静脉注射ACE2-/y小鼠建立高血压及心功能障碍模型,实验小鼠随机分为ACE2+/y对照组、ACE2-/y模型组、定心胶囊组(12.5g/kg)和卡托普利组(3.75mg/kg)。造模后,监测小鼠收缩压,检测心功能,观察信号转导及转录活化因子3(STAT3)和STAT1的表达及活化。结果:定心胶囊降低血浆Ang Ⅱ水平,显示持续的降压效果。心脏超声多普勒显示显著增加短轴缩短率和应变率、降低LVEDd和LVSDd并改善左室前壁厚度。蛋白免疫印迹显示定心胶囊降低STAT3的磷酸化并抑制其核转位。结论:定心胶囊改善Ang Ⅱ诱导的血压升高及心功能障碍,与其抑制STAT3的磷酸化及核转位有关。
Objective: To study the effect of Dingxin Capsule on blood pressure and cardiac dysfunction in ACE2- / y mice and its regulatory mechanism. Methods: ACE2- / y mice were injected with angiotensin Ⅱ (Ang Ⅱ) to establish models of hypertension and cardiac dysfunction. The experimental mice were randomly divided into ACE2 + / y control group, ACE2- / y model group, Dingxin capsule Group (12.5 g / kg) and captopril group (3.75 mg / kg). After modeling, systolic blood pressure was monitored, cardiac function was measured, and the expression and activation of signal transducer and activator of transcription 3 (STAT3) and STAT1 were observed. Results: Dingxin Capsule reduced plasma Ang Ⅱ level, showing sustained antihypertensive effect. Echocardiographic Doppler imaging significantly increased short axis shortening and strain rates, decreased LVEDd and LVSDd, and improved left ventricular anterior wall thickness. Western blotting showed that Dingxin capsule reduced the phosphorylation of STAT3 and inhibited its nuclear translocation. Conclusion: Dingxin Capsule can improve Ang Ⅱ - induced hypertension and cardiac dysfunction, which is related to the inhibition of phosphorylation and nuclear translocation of STAT3.
