目的研究Aurora-A高表达对人食管癌细胞的影响,以了解其在人食管癌侵袭转移中的作用。方法采用细胞转染的方法,利用Aurora-A基础水平表达较低的食管癌细胞系构建Aurora-A高表达细胞株,比较其与转染空载的细胞,分析二者在表型差别。结果Aurora-A高表达细胞表现出明显的增殖优势,在体外细胞移动实验中表现出穿膜能力大大增强,致瘤作用明显增强;提示Aurora-A高表达在人食管癌侵袭转移中有重要的意义。结论
Objective To study the effect of high expression of Aurora-A on human esophageal cancer cells to understand its role in invasion and metastasis of esophageal cancer. Methods Aurora-A high-expression cell line was constructed by using cell transfection method and using Aurora-A low-level esophageal cancer cell lines. The cells were transfected with no-load cells and their phenotypic differences were analyzed. Results The high expression of Aurora-A cells showed a significant proliferative advantage. In vitro cell migration experiments showed that the ability to penetrate the membrane was greatly enhanced and the tumorigenicity was significantly enhanced. This suggests that high expression of Aurora-A is important in the invasion and metastasis of human esophageal cancer. significance. in conclusion
