目的探讨大蒜素(Allicin)对皮肤基底细胞癌细胞放射治疗敏感性的影响及其与自噬水平的关系。方法采用MTT法检测Allicin对A431细胞的生长抑制作用,记录半数抑制浓度(IC50);将细胞分为4组:照射组(IR组)、Allicin组、IR+Allicin组以及对照组。细胞自噬水平的检测采用流式细胞仪。结果MTT检测结果显示,A431细胞经Allicin处理24,48,72h后,其活性受到显著的抑制,抑制程度受到时间及浓度的影响。同一浓度下,不同时间比较以及同一时间内不同浓度的比较,A431细胞活性的差异均具有统计学意义(P均<0.05)。Allicin作用于A431细胞24,48,72h的IC50分别为34.14,17.78,6.72mmol/L,差异均具有统计学意义(P均<0.05)。IR+Allicin组的SF_2,D_0,D_q分别为:75.81,6.46,1.34;IR组的SF_2,D_0,D_q分别为96.21,8.60,2.85。在各照射剂量点A431细胞的SF均显著低于IR组。与IR组比较,IR+Allicin组的肩区逐渐缩小,说明细胞的损伤修复能力呈现降低趋势。Allicin组和IR+Allicin组的A431细胞自噬水平均高于对照组,差异有统计学意义(P均<0.05)。结论 Allicin能上调人皮肤基底细胞癌A431细胞的自噬水平,进而增加A431细胞的放疗敏感性。
Objective To investigate the effect of Allicin on radiosensitivity of basal cell carcinoma of the skin and its relationship with autophagy. Methods The growth inhibition of Allicin on A431 cells was detected by MTT assay and the IC50 was recorded. The cells were divided into 4 groups: irradiation group (IR group), Allicin group, IR + Allicin group and control group. Detection of autophagy levels using flow cytometry. Results MTT assay showed that A431 cells treated with Allicin for 24, 48 and 72 hours significantly inhibited the activity of A431 cells and the degree of inhibition was affected by time and concentration. The same concentration, different time and different concentrations at the same time, A431 cell activity differences were statistically significant (P all <0.05). The IC50 of Allicin on A431 cells at 24,48,72h were 34.14,17.78,6.72mmol / L, respectively, with significant difference (all P <0.05). The SF_2, D_0 and D_q of IR + Allicin group were 75.81,6.46 and 1.34, respectively; SF_2, D_0 and D_q of IR group were 96.21, 8.60 and 2.85 respectively. The SF of cells in A431 cells at each irradiation dose point was significantly lower than that in IR group. Compared with the IR group, the shoulder area of the IR + Allicin group gradually decreased, indicating that the cell repair ability of injury showed a decreasing trend. Allicin group and IR + Allicin group A431 autophagy levels were higher than the control group, the difference was statistically significant (P all <0.05). Conclusion Allicin can up-regulate the autophagy of A431 cells in human skin and increase the radiosensitivity of A431 cells.
