建立了荷移荧光光谱法测定杨树桑黄提取物中总黄酮含量的方法。以芦丁为对照品,1%三氯化铝为受电子试剂,测定荷移复合物荧光强度,并系统考察了荷移反应条件对荧光强度的影响。芦丁在4—20μg·mL~(-1)浓度范围内线性关系良好,RSD为2.21%,平均回收率为100.3%。荷移反应后黄酮类物质产生荧光,可用于定量测定,方法简便、快速,适用于杨树桑黄总黄酮的含量测定。
A method was developed for the determination of total flavonoids in Poplar Phellodendri oil extract by charge-transfer fluorescence spectroscopy. Rutin was used as reference substance and 1% aluminum trichloride was used as electron-acceptor to measure the fluorescence intensity of charge-transfer complex. The influence of reaction conditions on fluorescence intensity was investigated systematically. Rutin had good linearity in the concentration range of 4-20μg · mL ~ (-1) with a RSD of 2.21% and an average recovery of 100.3%. Fluorescence reaction of flavonoids after the shift reaction can be used for quantitative determination, the method is simple and rapid, suitable for the determination of the total flavonoids content of Phellodendri mulberry.
