目的采用高压液相色谱法(HPLC)测定“黄芩-黄连”药对中黄芩4种有效成分(黄芩苷、汉黄芩苷、黄芩素、汉黄芩素)。方法采用Hypersil ODS2-C_(18)色谱柱(250 mm×4.6 mm,5μm);流动相为乙腈-0.3%磷酸三乙胺,梯度洗脱,流速:1.0 m L·min~(-1),检测波长:280 nm,柱温:30℃。结果黄芩苷、汉黄芩苷、黄芩素和汉黄芩素分别在0.048~0.288 mg·m L~(-1)、0.056~0.336 mg·m L~(-1)、0.042~0.254 mg·m L~(-1)和0.047~0.280mg·m L~(-1)范围线性关系良好,平均回收率分别为100.01%、99.99%、99.64%、99.79%,RSD分别为1.20%、0.80%、1.04%、0.94%。结论该方法简便,结果准确可靠,可作为“黄芩-黄连”药对提取物的质量控制标准。
OBJECTIVE To determine the four active ingredients (baicalin, baicalin, baicalein and wogonin) of “Scutellaria baicalensis and Coptidis Rhizoma” in Radix Scutellariae by high pressure liquid chromatography (HPLC). Methods Hypersil ODS2-C 18 column (250 mm × 4.6 mm, 5 μm) was used as the mobile phase. The mobile phase consisted of acetonitrile-0.3% triethylamine phosphate gradient with a flow rate of 1.0 mL · L -1, Detection wavelength: 280 nm, column temperature: 30 ℃. Results Baicalin, baicalin, baicalein and wogonin were in the range of 0.048 ~ 0.288 mg · m L -1, 0.056 ~ 0.336 mg · m L -1 and 0.042 ~ 0.254 mg · m L ~ (-1) and 0.047 ~ 0.280mg · m L -1, the average recoveries were 100.01%, 99.99%, 99.64% and 99.79%, respectively, and the RSDs were 1.20%, 0.80% and 1.04% , 0.94%. Conclusion The method is simple, accurate and reliable, and can be used as quality control standard of “Scutellaria baicalensis Georgi”.
