目的探讨抗肿瘤新药诺帝(Nordy)在体外对卵巢癌细胞(3AO黏液性癌,SKOV3浆液性癌)生长的抑制作用及对细胞周期的影响,同时对卵巢癌细胞株中Aurora-A的mRNA和蛋白过表达的抑制作用进行研究。方法MTT法检测诺帝对细胞增殖的抑制作用;RT-PCR和Westernblot检测Aurora-A的mRNA和蛋白表达情况;流式细胞术分析细胞周期。结果经诺帝作用后Aurora-A的mRNA及蛋白的表达水平显著下降(P<0.01);诺帝能明显抑制3AO、SKOV3细胞的增殖,阻滞细胞周期于G0/G1期(P<0.05)。结论诺帝对卵巢癌3AO、SKOV3细胞的生长具有明显抑制作用,可以阻滞细胞周期。在诺帝多环节的抑癌机制中,抑制癌细胞株Aurora-A的异常高表达,阻止中心体的异常转变可能是其机制之一。
Objective To investigate the inhibitory effect of a new antitumor drug Nordy on the growth of ovarian cancer cells (3AO mucinous carcinoma and SKOV3 serous carcinoma) and its effect on the cell cycle in vitro. Meanwhile, the expression of Aurora-A mRNA in ovarian cancer cell lines And inhibition of protein overexpression were studied. Methods MTT assay was used to detect the inhibition effect of Nordy on cell proliferation. The mRNA and protein expression of Aurora-A was detected by RT-PCR and Western blot. The cell cycle was analyzed by flow cytometry. RESULTS: Aurora-A mRNA and protein expression decreased significantly (P <0.01). Nordy significantly inhibited the proliferation of 3AO and SKOV3 cells and arrested the cell cycle at G0 / G1 phase (P <0.05) . Conclusion Nordy has a significant inhibitory effect on the growth of 3AO and SKOV3 ovarian cancer cells, which can block the cell cycle. In the Nordy multi-part of the tumor suppressor mechanism, inhibition of cancer cell lines Aurora-A abnormal high expression, blocking the abnormal changes in centrosome may be one of the mechanisms.
