目的:观察脑缺血及缺血再灌流海马CA1区 5-HT的变化,探讨脑缺血海马CA1区神经元损伤与 5-HT变化的关系。方法:用免疫组织化学ABC法及图像分析技术对沙土鼠脑缺血海马CA1区5-HT含量变化进行研究。结果:(1)脑缺血10 min时,沙土鼠海马CA1区5-HT免疫反应阳性纤维的平均光密度值(OD)与对照组比较无显著性差异,而缺血 30 min时,OD值则下降,4 h时下降得更多,P<0. 05。 (2)脑缺血 10 min再灌流30 min时 5-HT的 OD值即降低,再灌流 4 h时进一步降低,再灌流 24 h时最低,P<0.05,至 36 h时OD值逐渐恢复正常。结论:沙土鼠海马CA1区 5-HT含量在脑缺血及缺血再灌流时均降低,提示海马CA1区神经元缺血损伤可能与 5-HT的紊乱有关。
OBJECTIVE: To observe the changes of 5-HT in hippocampus CA1 area after cerebral ischemia and reperfusion, and to explore the relationship between neuronal damage and 5-HT in hippocampal CA1 area after cerebral ischemia. Methods: The changes of 5-HT content in hippocampus CA1 area of gerbils with cerebral ischemia were studied by immunohistochemical ABC method and image analysis technique. Results: (1) At 10 min of cerebral ischemia, the average optical density (OD) of 5-HT immunoreactive fibers in hippocampal CA1 area of gerbils was not significantly different from that of the control group, while at 30 min of ischemia, OD value Then decreased, dropping more at 4 h, P <0. 05. (2) The OD values of 5-HT decreased after reperfusion for 10 min at 10 min and further decreased at 4 h after reperfusion, and reached the lowest at 24 h after reperfusion Back to normal. CONCLUSION: The content of 5-HT in hippocampal CA1 of gerbils decreased in cerebral ischemia and ischemia-reperfusion, suggesting that ischemic injury of hippocampal CA1 neurons may be related to the disorder of 5-HT.
