目的 :探讨原癌基因C jun在大鼠变应性鼻炎鼻黏膜中的表达及其在变应性鼻炎发病机制中的作用。方法 :将 4 0只大鼠随机分为实验组 (30只 )和对照组 (10只 ) ,实验组根据制模成功后的时间又分为 30min组、4h组和 8h组 ,每组 10只。以卵清蛋白为致敏原建立大鼠变应性鼻炎动物模型 ,制模成功后 ,分别测各组动物的鼻分泌量 ;并取大鼠鼻黏膜组织 ,应用免疫组化技术、组织学检查等方法 ,检测C jun在鼻黏膜组织中的表达及各组鼻黏膜组织的病理学改变 ;应用计算机图像分析技术 ,测定阳性细胞的灰度值 ,并对鼻分泌量与阳性细胞灰度值进行相关分析。结果 :C jun免疫反应阳性物质呈棕色 ,分布于上皮细胞及多形核炎症细胞的细胞核中。30min组 ,阳性细胞数最多 ,其平均灰度值与对照组相比 ,差异有极显著性意义 (P <0 .0 1) ,并与鼻分泌量呈正相关 (r =0 .784 ,P <0 .0 5 ) ;4h组 ,C jun在炎症细胞内表达减弱 ,但其灰度值与对照组相比 ,差异仍有极显著性意义 (P <0 .0 1) ,亦与鼻分泌量呈正相关 (r =0 .6 2 6 ,P <0 .0 5 ) ;8h组 ,C jun的表达与对照组相比 ,差异无显著性意义 (P >0 .0 5 ) ,且与鼻分泌量无相关性。结论 :原癌基因C jun的表达可能是变应性鼻炎中炎症细胞活化的早期标志 ,与炎症细胞的增殖、分化有着密?
Objective: To investigate the expression of oncogene C jun in nasal mucosa of allergic rhinitis in rats and its role in the pathogenesis of allergic rhinitis. Methods: Forty rats were randomly divided into experimental group (30 rats) and control group (10 rats). The experimental group was divided into 30min group, 4h group and 8h group according to the time after successful modeling. . To ovalbumin as allergen to establish animal model of allergic rhinitis in rats, the successful model, the nasal secretion of each group of animals were measured; and nasal mucosa in rats, the application of immunohistochemistry, histological examination The expression of C jun in nasal mucosa and the pathological changes of nasal mucosa in each group were detected by computerized image analysis. The gray value of positive cells was determined by computer image analysis. The nasal secretion and gray value of positive cells related analysis. Results: C jun immunoreactive substance was brown and distributed in the nucleus of epithelial cells and polymorphonuclear inflammatory cells. 30min group, the highest number of positive cells, the average gray value compared with the control group, the difference was significant (P <0 01), and nasal secretion was positively correlated (r = 0.784, P < 0.05). Compared with the control group, the expression of C jun in the 4h group was weaker than that in the control group (P <0.01), and also correlated with the nasal secretion (R = 0.666, P <0.05). There was no significant difference in the expression of C jun between the 8h group and the control group (P> 0.05), and the positive correlation with nasal secretion No correlation Conclusion: The expression of C jun proto oncogene may be an early sign of activation of inflammatory cells in allergic rhinitis, which is closely related to the proliferation and differentiation of inflammatory cells.
