为了研究利什曼原虫(Leishmania)对于含五价锑药物的抗药性机理而建立了一种测定细胞内痕量金属锑的方法。本文尝试使用浓硝酸冷消化法处理细胞样品,简化了实验步骤,降低了被测物质损失和污染的可能性。电感耦合等离子体-质谱(ICP-MS)作为检测手段令本方法测定金属锑的最低检出浓度达到0.036μg?l-1,线性范围达到1.0-80ng?ml-1,加标回收率为98.0-110.0%,日间精密度≤1.63%。本方法较为简单,结果准确可靠,已经应用于寄生虫的金属抗药性研究,所获结果完全符合预期的实验模型。
In order to study the resistance mechanisms of Leishmania to pentavalent antimony-containing drugs, a method for the determination of trace antimony in cells was developed. This article attempts to use concentrated nitric acid cold digestion of cell samples, simplifying the experimental steps, reducing the possibility of material loss and contamination. Inductively coupled plasma-mass spectrometry (ICP-MS) was used as a detection method to determine the minimum detectable concentration of antimony metal in this method to 0.036μg? L-1 with a linear range of 1.0-80ng? Ml-1 and a recovery of 98.0 -110.0%, daytime precision≤1.63%. The method is relatively simple, the results are accurate and reliable, and has been applied to the study of metal resistance of parasites. The obtained results are completely in line with the expected experimental model.
