目的高效液相色谱(HPLC)法测定芹菜籽中3-正丁基苯酞的含量。方法采用YMC ODS C18柱(250 mm×4.6mm,5μm);流动相为乙腈-酯酸钠(45∶55),酯酸钠0.05 mol.L-1,冰酯酸调节pH4.6;检测波长为228 nm;流速为1.0ml.min-1;柱温:40℃。结果3-正丁基苯酞在0.044 1~0.161 7μg范围内线性关系良好,r=0.999 7,方法的回收率为98.19%,RSD为1.38%(n=5)。结论该方法能够准确可靠地测定3-正丁基苯酞的含量,可作为芹菜籽的质量控制指标之一。
Objective To determine the content of 3-n-butylphthalide in celery seed by high performance liquid chromatography (HPLC). Methods The YMC ODS C18 column (250 mm×4.6 mm, 5 μm) was used. The mobile phase consisted of acetonitrile-sodium(45:55) sodium aerylate and 0.05 mol.L-1 sodium citrate. The pH was 4.6. 228 nm; flow rate 1.0 ml.min-1; column temperature: 40°C. Results The linearity of 3-n-butylphthalide was good in the range of 0.044 1~0.161 7μg, r=0.999 7. The recovery of the method was 98.19%, and the RSD was 1.38% (n=5). Conclusion This method can accurately and reliably determine the content of 3-n-butylphthalein, which can be used as one of the quality control indexes of celery seed.
