硒是一种人体必需的微量元素。硒具有增强机体免疫功能的作用。本实验通过细胞毒性检测(MTT),凋亡细胞荧光染色计数法,流式细胞仪等方法,比较分析了免疫活性细胞(TIL和LAK细胞)在硒作用下,对人结肠癌细胞株(LoVo)的杀伤特点,并采用RT-PCR技术、原位杂交技术检测了上述免疫细胞和肿瘤细胞有关细胞凋亡信号传导基因Fas/FasL的表达情况。实验发现在一定硒浓度(10~(-6)~10~(-8)mol/L)条件下,可增强TIL和LAK细胞对LoVo细胞的杀伤活性,提高靶细胞的凋亡率。RT-PCR技术检测TIL细胞FasLm-RNA表达情况,发现经硒作用后,能提高TIL细胞的FasL的表达水平,而LoVo细胞则无明显变化。实验结果表明,硒在一定浓度下,可增强免疫活性细胞的抗癌作用,提高TIL细胞FasL和靶细胞的Fas表达水平,并通过Fas/FasL这一凋亡信号传导途径,杀伤表达Fas分子的靶细胞,促使其凋亡。

The experimental results show that selenium can increase the anti-cancer effect of immunocompetent cells at a certain concentration, increase the FasL and Fas expression levels of target cells, and kill the Fas molecules through Fas/FasL, apoptotic signal transduction pathway. The target cell promotes its apoptosis.
