目的了解具有家族背景大肠癌MMR基因(hMLH1、hMSH2)表达及细胞增殖活性特征。方法应用免疫组化SP法和流式细胞术(FCM),对46例大肠癌进行研究。结果具有家族背景大肠癌组织hMLH1表达阴性率及其切缘“正常”腺体hMLH1和hMSH2表达阴性率明显高于无家族史的散发性大肠癌(P<0.05)。而其PCNA标记指数、DNA异倍体率则显著减低(P<0.01,P<0.05)。结论具有家族背景大肠癌肿瘤细胞及切缘“正常”腺体hMLH1和hMSH2突变率的增高,其增殖指标有所降低。
Objective To understand the characteristics of MMR gene (hMLH1, hMSH2) expression and cell proliferation activity in colorectal cancer with family background. Methods Immunohistochemical SP method and flow cytometry (FCM) were used to study 46 cases of colorectal cancer. Results The negative rate of hMLH1 expression and the negative rate of hMLH1 and hMSH2 expression in colorectal cancer tissues with family background were significantly higher than that of sporadic colorectal cancer without family history (P<0.05). The PCNA index and DNA heteroploidy rate were significantly reduced (P<0.01, P<0.05). Conclusions The increased mutation rate of hMLH1 and hMSH2 with family background colorectal cancer cells and margin “normal” glands has decreased the proliferation index.
