目的:探讨复方中药对MRL小鼠心肌细胞凋亡的作用及机理。方法:将MRL小鼠分为复方中药组、桂枝组、强的松组和空白对照组,检测MRL小鼠心肌组织凋亡细胞,计算心肌细胞凋亡指数(AI),RT-PCR法检测心肌细胞凋亡基因Bax mRNA、Caspase 3 mRNA表达,观察复方中药(组成:丹参、桂枝、西洋参、冰片)对MRL小鼠心肌细胞凋亡的影响。结果:复方中药能抑制Bax mRNA、Caspase 3 mRNA表达,明显抑制心肌细胞凋亡(心肌细胞凋亡指数显著下降)。其疗效明显优于桂枝组和空白对照组(P<0.05),与强的松组疗效相当,P>0.05。结论:复方中药可能通过调控凋亡基因Bax mRNA、Caspase 3 mRNA表达而抑制心肌细胞凋亡,从而延缓或部分逆转MRL心肌损害的发生、发展。
Objective: To investigate the effect and mechanism of compound Chinese herbs on cardiomyocyte apoptosis in MRL mice. Methods: MRL mice were divided into compound Chinese herbal medicine group, Guizhi group, prednisone group and blank control group. Apoptotic cells were detected in myocardial tissue of MRL mice, and cardiomyocyte apoptosis index (AI) was calculated. RT- Effects of Compound Chinese Herbal Medicine (Composition: Salvia Miltiorrhiza, Guizhi, American Ginseng, Borneol) on Cardiomyocyte Apoptosis in MRL Mice Myocardial cell apoptosis gene Bax mRNA, Caspase 3 mRNA expression Results: Compound Chinese herbs can inhibit the expression of Bax mRNA and Caspase 3 mRNA, and significantly inhibit the apoptosis of cardiomyocytes (the apoptotic index of cardiomyocytes decreased significantly). The curative effect was obviously superior to the Guizhi group and the blank control group (P <0.05), which was similar to that of the prednisone group (P> 0.05). Conclusion: Compound Chinese herbal medicine may inhibit the apoptosis of cardiomyocytes by regulating the expression of Bax mRNA and Caspase 3 mRNA and thereby delay or partly reverse the occurrence and development of myocardial damage in MRL.
