目的观察牛膝醇提取物对去卵巢骨质疏松大鼠骨密度(BMD)、骨转换和骨组织Ⅰ型胶原蛋白表达的影响。方法背侧切口切除大鼠双侧卵巢,分为去卵巢组,牛膝提取物高、中、低剂量组和雌激素对照组,另设一假手术组,分别给予基础饲料和不同剂量治疗药物,12w后使用双能X线骨密度仪测量股骨、胫骨和腰椎的骨密度;采用酶联免疫吸附法对骨转换指标血清骨碱性磷酸酶(BAP)、血清骨钙素(BGP)、抗酒石酸酸性磷酸酶(TRACP)、尿Ⅰ型胶原氨基末端肽(NTx)/肌酐比值(NTx/Cr)进行测定,并利用Western免疫印迹法检测骨组织Ⅰ型胶原蛋白表达的变化。结果大鼠去势后骨密度和Ⅰ型胶原蛋白的表达显著下降(P<0.01),骨转换指标有较大变化。牛膝醇提取物高、中剂量组可使去势大鼠骨密度和Ⅰ型胶原蛋白的表达显著提高,改善骨转换指标的变化,且存在一定的剂量-效应关系。结论牛膝醇提取物能提高去势大鼠的骨密度和Ⅰ型胶原蛋白的表达。
Objective To investigate the effects of Achyranthes bidentata extract on bone mineral density (BMD), bone turnover and type Ⅰ collagen expression in ovariectomized rats. Methods Dorsal cut bilateral ovaries were divided into ovariectomized group, Achyranthes bidentata extract high, medium and low dose groups and estrogen control group, another set of a sham operation group, were given basal diet and different doses of drugs After 12 weeks, the bone mineral density of the femur, tibia and lumbar vertebrae were measured by dual-energy X-ray absorptiometry. Serum levels of bone alkaline phosphatase (BAP), serum osteocalcin (BGP) TRACP and NTx / creatin ratio (NTx / Cr) were measured. Western blotting was used to detect the expression of type Ⅰ collagen in bone tissue. Results The bone mineral density and the expression of type Ⅰ collagen decreased significantly after castration in rats (P <0.01), and the indexes of bone turnover changed greatly. Achyranthes bidentata alcohol extract high and medium dose group can significantly increase the bone mineral density and type Ⅰ collagen expression in ovariectomized rats to improve the change of bone turnover index, and there is a certain dose-response relationship. Conclusion Achyranthes bidentata extract can increase bone mineral density and type Ⅰ collagen protein in ovariectomized rats.
