目的探讨浸润相关基因KiSS-1和基质金属蛋白酶-9(MMP-9)在滋养细胞浸润调控中作用。方法采用逆转录聚合酶链反应技术和Western印迹法检测90例正常妊娠(早、中、晚期妊娠各30例)及40例子痫前期患者(15例轻度和25例重度)胎盘、30例葡萄胎、9例侵蚀性葡萄胎和8例绒毛膜癌组织中KiSS-1和MMP-9mRNA及其蛋白质的表达。结果妊娠早期MMP-9mRNA和蛋白质表达较高,前者吸光度(A)值为0·39±0·22,后者为(36±7)μg/100μg总蛋白质,随妊娠进展逐渐下降,至足月妊娠时表达显著低于妊娠早期(P均<0·01);而KiSS-1mRNA及蛋白质产物metastin则相反,随妊娠进展呈增高趋势(P均<0·01)。子痫前期MMP-9mRNA和蛋白质表达均显著低于妊娠晚期(P均<0·01),而KiSS-1mRNA和metastin表达则均与之相反。妊娠滋养细胞疾病组织中MMP-9基因和蛋白质的表达均显著高于早期妊娠(P均<0·01)。葡萄胎和侵蚀性葡萄胎组织中KiSS-1mRNA和metastin表达显著低于正常妊娠早期(P<0·05,P<0·01),绒毛膜癌组织中未见KiSS-1mRNA和metastin表达。结论促浸润基因MMP-9的表达变化与滋养细胞浸润活性呈正相关,而抑浸润基因KiSS-1的表达变化与之呈负相关,其相互作用在滋养细胞浸润活性调控中起重要作用。
Objective To investigate the role of infiltration-related genes KiSS-1 and MMP-9 in the regulation of trophoblast invasion. Methods 90 cases of normal pregnancy (30 cases of early, middle and late pregnancy) and 40 cases of preeclampsia (15 cases of mild and 25 cases of severe placenta) and 30 cases of grapevine were detected by reverse transcription polymerase chain reaction and Western blot, 9 cases of invasive hydatidiform mole and 8 cases of choriocarcinoma tissues and KiSS-1 and MMP-9 mRNA and protein expression. Results The expression of MMP-9 mRNA and protein in early gestation period was high. The absorbance (A) value of the former was 0.39 ± 0.22 and the latter was (36 ± 7) μg / 100μg total protein, and gradually decreased with the progress of pregnancy until term The expression in pregnancy was significantly lower than that in early pregnancy (P <0.01). KiSS-1 mRNA and protein metastin, on the contrary, increased with the progress of pregnancy (all P <0.01). The expression of MMP-9 mRNA and protein in preeclampsia was significantly lower than that in the third trimester of pregnancy (P <0.01), while the expressions of KiSS-1 mRNA and metastin were the opposite. The expression of MMP-9 gene and protein in gestational trophoblastic disease tissues were significantly higher than those in early pregnancy (all P <0.01). The expression of KiSS-1mRNA and metastin in hydatidiform mole and invasive hydatidiform mole were significantly lower than those in normal early pregnancy (P <0.05, P <0.01). KiSS-1mRNA and metastin expression were not found in choriocarcinoma. Conclusions There is a positive correlation between the expression of MMP-9 and the infiltration of trophoblast while the expression of KiSS-1 is negatively correlated. The interaction plays an important role in the regulation of trophoblast invasion.
