目的观察4,5,7-三羟基异黄酮(Genistein)和阿霉素(ADM)单用和联用对诱导体外培养的人舌癌细胞株Tca8113凋亡的影响。方法以体外培养的人舌癌细胞株Tca8113为实验模型,Genistein与ADM联合作用于癌细胞,倒置相差显微镜下观察药物处理后癌细胞的细胞形态学变化,四甲基偶氮唑蓝比色法分析药物对癌细胞的增殖抑制作用,流式细胞仪测定细胞凋亡率和细胞周期分布。结果Genistein和ADM两者单用对癌细胞均有生长抑制作用,且随剂量升高抑制趋势增强(P<0.05)。两药联用对癌细胞的抑制作用优于两者的单纯相加(Q>1)。联合用药使肿瘤细胞S期延长,G2/G1值缩小,凋亡率明显升高(P<0.05)。结论Genistein与ADM联用对Tca8113细胞具有协同抑制作用,其作用机理可能是联合用药促使肿瘤细胞S期延长,G2/G1值缩小,提高了ADM的诱导凋亡效应。
Objective To investigate the effects of genistein and doxorubicin (ADM) on the apoptosis of human tongue cancer cell line Tca8113 cultured in vitro. Methods Human tongue cancer cell line Tca8113 cultured in vitro was used as an experimental model. Genistein and ADM were used to treat cancer cells. The morphological changes of cancer cells were observed under inverted phase contrast microscope. Methyl thiazolyl tetrazolium The inhibitory effect of drugs on the proliferation of cancer cells was analyzed. The apoptosis rate and cell cycle distribution were determined by flow cytometry. Results Both Genistein and ADM had inhibitory effects on the growth of cancer cells alone. The inhibition of genistein and ADM increased with the increase of dosage (P <0.05). The combined effect of the two drugs on cancer cells was better than the simple addition (Q> 1). The combination of drugs prolonged S phase of tumor cells, reduced G2 / G1 value and significantly increased the apoptosis rate (P <0.05). Conclusions Genistein combined with ADM has a synergistic inhibitory effect on Tca8113 cells. The mechanism may be that the combination of Genistein and ADM can prolong the S phase and reduce the G2 / G1 value of the tumor cells, and improve the induction of apoptosis of ADM cells.
