目的:应用单纯胶原酶法、复合胶原酶法培养大鼠血管平滑肌细胞,比较其生长特性。方法:采用改良Lowry法测定了蛋白质含量,依照荧光指示剂法测定了细胞内[Ca2+]。结果:2种方法其生长曲线类似,均于7~8d至高峰,其蛋白质含量略有差异,也是在7~8d达最大,钙含量分别为(202.60±15.18)nmol/L和(153.20±11.34)nmol/L。结论:单纯胶原酶法相对经济,复合胶原酶法能更好地保护细胞,可根据具体情况选用。
OBJECTIVE: To study the growth characteristics of rat vascular smooth muscle cells by collagenase method and collagenase method. Methods: The content of protein was determined by modified Lowry method and the intracellular [Ca2 +] was determined by fluorescence indicator method. Results: The growth curves of the two methods were similar and all reached the peak from 7 to 8 days with a slight difference in protein content, reaching the maximum at 7-8 days with the calcium content of (202.60 ± 15.18) nmol / L and (153.20 ± 11.34) nmol / L. Conclusion: Simple collagenase method is relatively economical, complex collagenase method can better protect cells, can be selected according to the specific circumstances.
