目的研究奥氮平与氟西汀单独或两药联合给药对大鼠脑内柠檬酸合成酶活性短期和长期的影响。方法 135只Wistar大鼠随机分为对照组与实验组。对照组腹腔注射生理盐水,实验组再分为几个亚组。2个剂量奥氮平组(3,6 mg·kg~(-1)),2个剂量氟西汀组(12.5,25.0 mg·kg~(-1)),联合用药组:A组(3 mg·kg~(-1)奥氮平+12.5 mg·kg~(-1)氟西汀)、B组(3 mg·kg~(-1)奥氮平+25.0 mg·kg~(-1)氟西汀)、C组(6 mg·kg~(-1)奥氮平+12.5 mg·kg~(-1)氟西汀)、D组(6 mg·kg~(-1)奥氮平+25.0 mg·kg~(-1)氟西汀),连续给药28 d。用分光光度法测定并比较第1次给药后2 h、末次给药后2,24 h的大鼠前额叶皮层、海马区和纹状体柠檬酸合成酶的活性。结果与对照组相比,在给药第1次后2 h,2个剂量奥氮平组、大剂量氟西汀组、联合A组的大鼠海马区柠檬酸合成酶活性明显增加(P<0.05)。结论短程小剂量奥氮平联合氟西汀可显著增加大鼠脑内柠檬酸合成酶活性。
Objective To study the short-term and long-term effects of olanzapine and fluoxetine on the activity of citrate synthase in rat brain. Methods 135 Wistar rats were randomly divided into control group and experimental group. Control group, intraperitoneal injection of saline, the experimental group is divided into several subgroups. Two doses of olanzapine (3, 6 mg · kg -1) and two doses of fluoxetine (12.5 mg · kg -1, 25.0 mg · kg -1) (0 mg · kg -1 olanzapine +12.5 mg · kg -1 fluoxetine), group B (3 mg · kg -1 olanzapine +25.0 mg · kg -1 ) Fluoxetine, group C (6 mg · kg -1 olanzapine +12.5 mg · kg -1 fluoxetine), group D (6 mg · kg -1 olanzapine) Flat +25.0 mg · kg -1 fluoxetine) for 28 days. The activity of citrate synthase in rat prefrontal cortex, hippocampus and striatum was determined and compared by spectrophotometry at 2 h after the first administration and at 2,24 h after the last administration. Results Compared with the control group, the activity of citrate synthase in hippocampus of rats in two doses olanzapine group, high-dose fluoxetine group and group A was significantly increased 2 h after the first administration (P < 0.05). Conclusion Short-term and low-dose olanzapine combined with fluoxetine can significantly increase the activity of citrate synthase in rat brain.
