目的:观察白英水提物对胃癌SGC-7901细胞凋亡及Notch1基因表达的影响。方法:常规法制备白英水提物,实验分正常对照组、白英处理组。白英处理组加入白英水提物终浓度分别为12.5、25mg/mL和50mg/mL。MTT法检测细胞增殖抑制率,流式细胞仪检测细胞凋亡,半定量RT-PCR和免疫化学分析细胞中Notch1、caspase-9基因表达。结果:与正常对照组比较,白英处理组SGC-7901细胞增殖有显著性下降,细胞凋亡显著性增多(P<0.05),Notch1基因表达呈显著性的下降(P<0.05)、caspase-9基因表达有明显的上调(P<0.05),并随白英浓度增加而加强。结论:白英水提物能下调人胃癌SGC-7901细胞中Notch1信号蛋白表达和促进细胞凋亡,这可能是其发挥抗癌作用的重要机制之一。
Objective: To observe the effects of aqueous extract from Baiying on the apoptosis of gastric cancer cell line SGC-7901 and the expression of Notch1 gene. Methods: Conventional preparation of white water extract, the experimental points normal control group, white British treatment group. Baiying treatment group to join the white water extract final concentrations were 12.5,25 mg / mL and 50mg / mL. The inhibitory rate of cell proliferation was detected by MTT assay. Apoptosis was detected by flow cytometry. Semiquantitative RT-PCR and immunohistochemistry were used to analyze the expression of Notch1 and caspase-9. Results: Compared with the normal control group, the proliferation of SGC-7901 cells in Baiying treatment group was significantly decreased (P <0.05), the expression of Notch1 gene was significantly decreased (P <0.05), while the expression of caspase- 9 gene expression was significantly increased (P <0.05), and with the concentration of Baiying increased. CONCLUSION: Baiying water extract can down-regulate the expression of Notch1 protein and promote cell apoptosis in human gastric cancer cell line SGC-7901, which may be one of its important anti-cancer mechanisms.
