甲酰四氢叶酸钙(CF)一直是作为抗癌药MTX的解毒救援剂,本身并无抗癌活性。但文献报道,细胞内高浓度的CF,可增加5-Fu抑制脱氧胸苷酸合成酶活性。近年临床研究提示,CF能增高5-Fu对胃肠道恶性肿瘤的疗效;但也有部分临床实验结果表明并无增效作用。因此,CF对5-Fu抗癌活性影响究竟如何目前尚无定论。我们采用体外细胞实验方法,以人胃癌细胞SGC-7901为材料,
Formyl tetrahydrofolate (CF) has been used as a detoxification rescue agent for the anticancer agent MTX, and has no anticancer activity by itself. However, it has been reported in the literature that high concentrations of CF in cells can increase 5-Fu’s inhibition of deoxythymidylate synthase activity. Clinical studies in recent years have suggested that CF can increase the efficacy of 5-Fu in the treatment of gastrointestinal malignancies; however, some clinical trials have shown no synergistic effect. Therefore, the impact of CF on the anti-cancer activity of 5-Fu has not yet been determined. We used in vitro cell assays and human gastric cancer cells SGC-7901 as material.
