目的了解树突细胞(DCs)在类风湿关节炎(RA)发病机制中的作用,对RA滑膜液是否能影响DCs的活化状态进行了研究。方法体外诱导外周血单个核细胞转化成为非活化的DCs,用此诱导的DCs再经RA滑膜液培养48h,然后用流式细胞仪对细胞表面表达的人类白细胞抗原(HLA)-DR和共刺激分子CD80进行分析。结果经滑膜液培养的DCs表达CD80为(45±13)%高于对照组的(26±6)%,差异有统计学意义;经滑膜液培养的DCs表达HLA-DR为(67±16)%高于对照组的(60±4)%,差异无统计学意义。结论 RA滑膜液在体外能诱导DCs活化。
Objective To investigate the role of dendritic cells (DCs) in the pathogenesis of rheumatoid arthritis (RA) and investigate whether RA synovial fluid can affect the activation status of DCs. Methods Peripheral blood mononuclear cells (DCs) were induced to transform into non-activated DCs in vitro. DCs induced by DCs were cultured in RA synovial fluid for 48h. Then the human leukocyte antigen (HLA) -DR Stimulation molecule CD80 was analyzed. Results The expression of CD80 in DCs cultured in synovial fluid was (45 ± 13)% higher than that in control group (26 ± 6)%, and the difference was statistically significant. The expression of HLA-DR in DCs cultured in synovial fluid was (67 ± 16)% higher than the control group (60 ± 4)%, the difference was not statistically significant. Conclusions RA synovial fluid can induce DCs activation in vitro.
