目的通过检测游离肉碱及酰基肉碱在多囊卵巢综合征患者血清中的表达水平,探讨其在多囊卵巢综合征的发生机制及其作用。方法本研究纳入40例多囊卵巢综合征患者作为实验组,29例正常女性作为对照组。通过NeoBaseTMNon-derivatized MSMS试剂盒及串联质谱(MSMS)系统进行检测分析实验组和对照组血清样本中游离肉碱及酰基肉碱的表达水平。结果游离肉碱在两组中的表达水平差异无统计学意义。多囊卵巢综合征患者中甲基巴豆酰基肉碱、戊二酰基肉碱、己二酰基肉碱等12种酰基肉碱的表达水平均低于对照组,差异有统计学意义(P<0.05),而乙酰基肉碱在多囊卵巢综合征患者中表达升高,差异有统计学意义(P<0.05)。结论肉碱在多囊卵巢综合征患者血清中存在表达异常,乙酰基肉碱在多囊卵巢综合征患者血清中表达升高,酰基肉碱的异常表达可能参与多囊卵巢综合征的发生发展,且与胰岛素抵抗及内分泌异常相关。
Objective To detect the expression of free carnitine and acylcarnitine in the serum of patients with polycystic ovary syndrome and to explore its mechanism and its role in polycystic ovary syndrome. Methods The study included 40 patients with PCOS as experimental group and 29 normal women as control group. The expression levels of free carnitine and acylcarnitine in the serum samples of experimental group and control group were detected by NeoBaseTMNon-derivatized MSMS kit and tandem mass spectrometry (MSMS) system. Results There was no significant difference in the expression level of free carnitine between the two groups. The expression level of 12 kinds of acylcarnitine, such as methyl-crotonyl carnitine, glutaryl carnitine and adipoyl carnitine in patients with polycystic ovary syndrome was lower than that of the control group (P <0.05) , While acetyl carnitine in patients with polycystic ovary syndrome increased expression, the difference was statistically significant (P <0.05). Conclusions Carnitine is abnormally expressed in the serum of patients with PCOS. Acetylglucosamine is highly expressed in the serum of PCOS patients. The abnormal expression of acylcarnitine may be involved in the occurrence and development of polycystic ovary syndrome. And with insulin resistance and endocrine abnormalities related.
