目的研究细胞周期素依赖性激酶8(CDK8)及转化生长因子-β1(TGF-β1)在结直肠癌组织中的表达及与其临床病理因素的相关性。方法采用荧光定量PCR及免疫组化方法检测40例结直肠癌组织及其癌旁组织中CDK8和TGF-β1的mRNA及蛋白表达,以及它们在结直肠癌组织中表达的相关性。结果结直肠癌组织中CDK8和TGF-β1的mRNA表达水平显著高于癌旁组织(P<0.05);CDK8 mRNA的表达水平与结直肠癌淋巴结转移以及分期和肿瘤浸润有关(P<0.05),与患者年龄、性别、肿瘤大小及分化程度无关(P>0.05)。TGF-β1的mRNA的表达水平与结直肠癌淋巴结转移以及分期、浸润程度和肿瘤大小相关(P<0.05),与患者年龄、性别和肿瘤分化程度无关(P>0.05)。结直肠癌组织中CDK8 mRNA及TGF-β1 mRNA表达呈正相关关系(r=0.387,P=0.048)。结论CDK8和TGF-β1表达上调可能与结直肠癌的发生发展相关,且二者存在协同作用,可能与结直肠癌生物学行为及预后有关。
Objective To investigate the expression of cyclin-dependent kinase 8 (CDK8) and transforming growth factor-β1 (TGF-β1) in colorectal cancer and its correlation with clinicopathological parameters. Methods The mRNA and protein expressions of CDK8 and TGF-β1 in 40 colorectal cancer tissues and adjacent normal tissues were detected by fluorescence quantitative PCR and immunohistochemistry. Their correlations were also analyzed in colorectal cancer tissues. Results The mRNA expression of CDK8 and TGF-β1 in colorectal cancer tissues was significantly higher than that in adjacent non-cancerous tissues (P <0.05). The expression of CDK8 mRNA was correlated with the lymph node metastasis and stage and tumor infiltration in colorectal cancer (P <0.05) No correlation was found with age, sex, tumor size and differentiation (P> 0.05). The mRNA expression of TGF-β1 was correlated with lymph node metastasis, staging, infiltration and tumor size in colorectal cancer (P <0.05), but not with age, sex and tumor differentiation (P> 0.05). There was a positive correlation between CDK8 mRNA and TGF-β1 mRNA expression in colorectal cancer tissues (r = 0.387, P = 0.048). Conclusion The up-regulation of CDK8 and TGF-β1 may be related to the occurrence and development of colorectal cancer, and their synergistic effects may be related to the biological behavior and prognosis of colorectal cancer.
