目的:研究肿瘤抑制基因BRCA1对胰腺癌的作用和其蛋白在胰腺癌组织中的分布和肿瘤发生中的蛋白特性.方法:以乳腺癌的细胞株作为阳性对照组,对胰腺癌组织中的BRCA1亚细胞分布做了免疫荧光双重染色,免疫组织化学等实验并进行量化.结果:BRCA1蛋白在胰腺癌的组织中呈一定比例分布,这点在不同的胰腺癌患者组织标本中得到证实.免疫荧光双重染色法证实BRCA1蛋白在胰腺癌细胞胞质和胞核间互动,这是国内首次发现和报道BRCA1在胰腺癌组织细胞中胞质和胞核间互动现象和其亚细胞分布情况.结论:我们的多病例研究结果都证实BRCA1的胞质胞核有比例明确分布,提示可能是参与胰腺癌组织中蛋白功能调节机制,并可能具有与胰腺癌发生相关的蛋白特性,在胰腺癌细胞其亚细胞分布现象预示着将有更好的胰腺癌临床治疗的生物靶点.
OBJECTIVE: To study the effect of tumor suppressor gene BRCA1 on pancreatic cancer and the distribution of its protein in pancreatic cancer tissue and the characteristics of tumorigenesis.Methods: Breast cancer cell line was used as a positive control group to detect the expression of BRCA1 Subcellular distribution was done by immunofluorescence double staining, immunohistochemistry and other experiments and quantified.Results: BRCA1 protein in pancreatic cancer tissue showed a certain percentage of distribution, which is confirmed in different pancreatic cancer tissue samples.Immunofluorescence Dual staining confirmed the interaction of BRCA1 protein in the cytoplasm and nucleus of pancreatic cancer cells, which is the first time to discover and report the interaction between BRCA1 and the cytoplasm and nucleus in pancreatic cancer cells and its subcellular distribution.Conclusion: The results of multiple case studies confirm that BRCA1 has a well-proportioned distribution of cytoplasmic nuclei, suggesting that it may be involved in the regulation of protein function in pancreatic cancer tissues and may have the characteristics of proteins related to the development of pancreatic cancer. Distribution indicates that there will be a better clinical target for pancreatic cancer.
