目的 探索 HSV1- TK基因联合应用无环鸟苷(ACV)对人肾癌细胞株 GRC- 1细胞的杀伤作用 .方法 对分别培养及混合培养的 GRC- 1细胞和 GRC- 1/ TK细胞 ,用不同浓度 ACV进行处理 ,然后观察细胞形态及数量的改变 .结果 在 ACV作用下 ,GRC- 1细胞的形态及数量无明显改变 ;GRC- 1/ TK细胞被明显杀伤 ,数量明显减少 ,当含 2 5 .0 %GRC- 1/ TK细胞的混合培养细胞即可观察到明显杀伤作用 .结论 HSV1- TK基因转染联合应用无环鸟苷对人肾癌细胞株 GRC- 1细胞具有明显的杀伤作用 ,并存在较强的“旁观者效应”.
Objective To explore the cytotoxicity of HSV1-TK gene combined with acyclovir (ACV) on human renal cancer cell line GRC-1 cells.Methods GRC-1 cells and GRC-1 / TK cells were cultured and mixed with Then treated with different concentrations of ACV, and then observed the changes of cell morphology and number.Results GRC-1 cells in the presence of ACV had no significant changes in the morphology and number of cells; GRC-1 / TK cells were significantly killed, the number was significantly reduced, 5% GRC-1 / TK cells could be observed.Conclusion HSV1-TK gene transfection combined with acyclovir has a significant killing effect on human renal cancer cell line GRC-1 cells , And there is a strong “bystander effect”.
