目的研究胃癌组织、异型增生组织和正常胃粘膜组织中Aurora-A和TACC3的表达水平及其临床病理意义。方法采用免疫组化检测胃癌、异型增生组织和正常胃粘膜中Aurora-A、TACC3的表达,通过Log-rank检验、Cox多因素分析,揭示Aurora-A、TACC3的表达与胃癌生物学行为的关系。结果正常胃粘膜组织中Aurora-A蛋白均为阴性;异型增生组织中阳性率50.0%;而在胃癌组织中阳性率74.0%,正常胃粘膜组织中TACC3阳性率为4.16%;TACC3在异型增生组织阳性率为40.0%;癌组织中阳性率为64.00%;Aurora-A和TACC3在进展期胃癌中表达率分别为78.57%、69.0%,在早期胃癌的表达率分别为50%、37.5%;Aurora-A、TACC3的阳性表达与肿瘤的分化程度、是否伴有淋巴结转移显著相关,差异均有统计学意义(P<0.05),分化程度越低,Aurora-A、TACC3表达越高;有淋巴结转移的胃癌组织中Aurora-A、TACC3表达越强;生存曲线分析显示Aurora-A及TACC3阳性的胃癌患者生存时间比表达阴性组短,差异有统计学意义(P<0.05)。单因素Log-rank检验表示,Aurora-A,TACC3阳性表达对患者生存率具有影响,相对危险度分别为2.35、2.01。结论 Aurora-A和TACC3蛋白参与胃癌细胞的发生、发展,可以作为胃癌恶性程度及预后的评价指标
Objective To study the expression of Aurora-A and TACC3 in gastric cancer, dysplasia and normal gastric mucosa and their clinicopathological significance. Methods The expression of Aurora-A and TACC3 in gastric cancer, dysplasia tissues and normal gastric mucosa were detected by immunohistochemistry. Log-rank test and Cox multivariate analysis revealed the relationship between the expression of Aurora-A and TACC3 and the biological behavior of gastric cancer . Results The positive rate of Aurora-A protein in normal gastric mucosa was all negative; the positive rate in dysplasia was 50.0%; while the positive rate in gastric cancer was 74.0%, the positive rate of TACC3 in normal gastric mucosa was 4.16%; TACC3 in dysplasia The positive rate was 40.0%, the positive rate was 64.00%. The expression rates of Aurora-A and TACC3 in advanced gastric cancer were 78.57% and 69.0%, respectively. The rates of expression of Aurora-A and TACC3 in early gastric cancer were 50% and 37.5% The positive expression of -A and TACC3 was significantly correlated with the degree of tumor differentiation and lymph node metastasis (P <0.05). The lower the degree of differentiation was, the higher the expression of Aurora-A and TACC3 was. The positive rate of lymph node metastasis The survival curves of Aurora-A and TACC3 were higher in gastric cancer tissues than those in the negative expression group (P <0.05). Survival curve analysis showed that the survival time of Aurora-A and TACC3-positive gastric cancer patients was shorter than that of negative ones. Single-factor Log-rank test showed that the positive expression of Aurora-A and TACC3 had an impact on the survival rate of patients with relative risk of 2.35,2.01 respectively. Conclusion Aurora-A and TACC3 are involved in the occurrence and development of gastric cancer cells, which can be used as an indicator of malignancy and prognosis of gastric cancer
