Kupffer细胞(KC)是存在于肝窦状隙内表面的巨噬细胞,占机体组织巨噬细胞的80%~90%,是肝脏发挥解毒、抗瘤作用的主要效应细胞.数十年来,对KC的结构及直接抗癌作用已进行了广泛研究,而对KC在介导抗原特异性免疫反应尤其是将其作为抗原递呈细胞的研究尚不多见,可能与KC分离技术困难和对其生物学性质认识不足有关.我们采用0.05%胶原酶与0.1%链酶蛋白酶E联合门脉灌注及体外消化方法成功获得了具有相当数量和免疫活性的KC,在此基础上初步探讨了CD40/CD40L分子对KC抗原提呈功能的影响.
Kupffer cells (KC) are macrophages that exist in the inner surface of sinusoids, accounting for 80% -90% of the macrophages in the body tissue. They are the main effector cells for detoxification and anti-tumor effect of the liver. For decades, The structure and direct anticancer effect of KC have been extensively studied. However, there are few studies on KC that mediate antigen-specific immune responses, especially as antigen-presenting cells, which may be difficult to separate from KC Biological properties of lack of understanding.We use 0.05% collagenase and 0.1% Streptzyme E combined with portal vein perfusion and in vitro digestion successfully obtained a considerable amount and immunological activity of KC, based on the preliminary study of CD40 / CD40L Effect of Molecule on KC Antigen Presentation Function.
